Rohu (Labeo rohita) CuZn superoxide dismutase cDNA clone in expression system for recombinant protein-polyclonal antibody-ELISA based assay

Background:
Superoxide dismutases (SODs) comprise a large family of enzymatic antioxidants which are well-known metalloenzymes that catalyze the dis-mutation of superoxide into oxygen and hydrogen peroxide. CuZnSODs are found in the cytosol and extracellular compartments of eukaryotic cells and in the periplasm of Gram-negative bacteria. This group is generally considered as the most important group of SOD because of their physiological and therapeutic importance. Since the level of CuZnSOD gene expression varies in aquatic eco-systems following metal contamination, it can be used as an excellent biomarker in ecotoxicology and bioaccumulation studies. However, lack of availability of species specific proteins and assay system pose a serious concern in its estimation or further analysis in different stressful conditions in fish. Technology Details:
Rohu CuZnSOD was found to be having strong antioxidant activity. It’s presence in serum at low level could be easily detected by standardized ELISA using this produced polyclonal antibody and the recombinant protein generated in this technology, for its further evaluation as marker in ecotoxicological studies.
